RTK-RAS pathway. a Image showing the activation of the MAPK transcription factor by binding of growth factor to a transmembrane receptor tyrosine kinase. The resulting signaling cascade culminates with translocation of ERK/MAPK to the nucleus, where ERK activates transcription factors that ultimately result in cell growth. BRAF inhibitors, such as dabrafenib, block RAF signal and MEK/MAPK inhibitors, such as trametinib, block the MEK/MAPK signal. ad Contrast-enhanced CT images of the chest of a 52-year-old woman with BRAF V600 mutant melanoma, progressed on ipilimumab, treated with dabrafenib and trametinib. Baseline (a, b) and follow-up CT (c, d) performed two months after treatment was started, shows almost complete resolution of multiple lung nodules and mediastinal masses
